0,05 g sampel daging ayam
Tambahkan 1g Selenium dan 3 tetes H2O2 dan 2,5 ml H2SO4 pekat
Dimasukkan dalam tabung reaksi
Destruksi di ruang asam hingga jernih
didinginkan
Diencerkan dengan H2O 50 ml aquades, dikocok masukkan ke labu kjehldal
Didestilasi
Sampel yang diencerkan dengan aquades dimasukkan 10 ml ke labu kjehldal
Tambahkan penolphtalein 3 tetes dan NaoH 50% sampai larutan menjadi merah
Destilat ditampung dalam erlenmeyer berisi
5 mL Asam Borat 3%, aquades 25 ml dan 2 tetes indicator hingga larutan yang
mengandung uap Nitrogen menjadi 150 ml
Destilat dititrasi dengan HCl 0,01 N
Kadar Protein dihitung dengan blanko 0,05

50 ml Dipasangkan pada alat ekstraksi soxhlet, dihidupkan
Diamati perubahan warna, selama perebusan sampel
Setelah jernih alat ekstraksi dimatikan
Bahan direbus, sampai labu boiling tampak jernih
atau sampai lemak tidak terlihat lagi
Dicatat hasilnya
Sampel dikeringkan dalam oven 105 C selama 12 jam
