AUSTRAK
IODINASI (1-125) FSH MENGGUNAKAN OKSIDATOR N-BROMO SUKSINIMID,
LAKTOPEROKSIDASE DAN IODOGEN. FSH 1251adalah salah satu pereaksi Kit RIAyang
digunakan untuk menentukan konsentrasi FSH di dalam serum darah serta mengetahui
penyimpan~an koordinasi kelenjar hipotalamus, pituitari dan gonad. Untuk dapat mempro
duksi FSH1 51yang bermutu baik sebagai komponen pereaksi Kit RIAFSH, dalam penelitian
ini telah dilakukan iodinasi FSH dengan 1-125,menggunakan oksidator N-bromo suksinimid,
laktoperoksidase, dan iodo bead sebagai aseptor elektron :

Terhadap masing-masing hasil penandaan dilakukan penentuan penandaan dan uji imuno
logi serta ikatan non spesifik (NSB) dengan membandingkannya dengan persen

(Btr) Kit RIAFSH dari Amersham. Stabilitas ikatannya dalam penyimpanan pada 4 c juga
ditentukan. Dengan oksidator N-bromo suksinimid, lakto peroksidase dan iodogen diperoleh
hasil berturut-turut : persen penandaan 25 ,8,2 dan 18 ,nilai Btrsebesar 22 ,34
dan 25 serta NSB masing-masing sebesar 1,3; 2,3 dan 0,6 .Uji stabilitas menunjukkan
bahwa FSH_1251masih baik pada penyimpanan selama satu bulan pada suhu 4C.

THE USE OF N-BROMOSUCCINIMIDE, LACTOPEROXIDASEAND IODOGEN FOR

RIA kit reagent utilize for determination of FSH concentration in blood serum, to study the
coordinationdisorder of the principle hormones, i.e., hypothalamus, pituitary and gonads. To

(1-125)of FSH employing mild oxidant N-bromosuccinimid, lactoperoxidase, and iodo-bead
has been performed.

The percentage oflabeling, the immunology activity and Non Spesific Binding ofthe labeled
products were determined. The immunology activity in ) T and non spesific binding )
NSB were compare with Amersham FSH RIAkit. The stability ofthe binding is also be checked
by storing the labeled products at 4C. With N-bromosuccinimid, lactoperoxidase and iodogen
as the oxidizing agents, the results obtained were respectively as follows : percentages of
labelling were 25 ,8.2 and 18 . T values were 22 ,34 and 25 ,and the NSB's
were 1.3, 2.6 and 0.6 .The stability test of labeled FSH.1251showed that it does not
deteriorate significantly after storage for one month at 4C.
