The advantages of coating procedures that do merit careful consideration:

1. The supply of a variety of reliable-quality excipients, which are acceptable to regulatory agencies.

2. A successful track record of products being taken from research and development through pilot-scale to large-scale manufacturing with, in many cases, surprisingly few problems.

3. Manufacturing methods that normally do not require the fabrication of any new manufacturing equipment but rely on well-tested manufacturing methods, which in many instances, are readily validated.

4. The availability of a remarkably flexible range of formulation and processing variables, which can be used to custom build controlled release profiles for a plethora of purposes.

The three main functions of materials used as controlled-release coatings:

1. provisions of the backbone structure of the coating.

2. facilitation or control over transport of drug across the membrane.

3. plasticization of the coating.

A plasticizer : is an excipient usually low molecular weight organic solvent, added to coating material to assist in the processing by rendering the film more flexible.

It is believed that such materials act by lowering the polymer glass-transition temperature.

Plasticizers must be compatible, in terms of solubility parameter, with the polymer to which they are added to make it more flexible. Thus, plasticizers are not general purpose but must be chosen from materials that have been shown to be useful for a particular polymer.

Bioavailability study requirements for controlled-release products

The aims of these requirements are to determine that the following conditions are met:

1. The drug product meets the controlled-release claims made for it.

2. The bioavailability profile established for the drug product rules out the occurrence of clinically significant dose dumping. This is usually achieved by the conduct of a food effect study whereby the drug is administered with and without a high-fat breakfast.

3. The drug product’s steady-state performance is equivalent to a currently marketed non-controlled-or controlled-release drug product that contains the same active drug ingredient or therapeutic moiety and that is subject to an approved full new drug application.

4. The drug product’s formulation provides consistent pharmacokinetic performance between individual dosage units.

The reference material for such a bioavailability study shall be chosen to permit an appropriate scientific evaluation of the controlled-release claims made for the drug product.

The reference material is normally one of the following:

1. A solution or suspension of the active drug ingredient or therapeutic moiety.

2. A currently marketed immediate-release drug product containing the same active drug ingredient or therapeutic moiety and administered according to the dosage recommendations in the labeling of immediate-release drug product.

3. A currently marketed controlled-release drug product subject to an approved full new drug application containing the same active drug ingredient or therapeutic moiety and administered according to the dosage recommendations in the labeling proposed for the controlled-release drug product.

Dissolution Testing for Controlled-Release Drug Products

In vitro dissolution testing is important for:

1. providing process control and quality assurance

2. determining stable release characteristics of the product over time

3. facilitating certain regulatory determinations (e.g., absence of effect of minor formulation changes or of change in manufacturing site on performance)

4. in certain cases, especially for oral controlled-release formulations, the dissolution test, related to an in vivo/ in vitro correlation (IVIVC), can serve not only as a quality control for the manufacturing process but also as an indicator of how the formulation will perform in vivo.

5. A main objective of developing and evaluating an IVIVC is to establish the dissolution test as a surrogate for human bio-equivalence studies.
