Penuaan Sel-Karena aktifitas sel menurun
-Stress oksidatif di dalam sel merupakan penyebab
proses aging
-Mitokondria yang menghasilkan ROS (reactive oxygen spescies) memegang peranan dalam proses aging suatu organisma
- Superoksida dgn konsentrasi tertentu dapat menurunkan daya tahan hidup organisme invertebrata
-Pada mamalia, pemberian antioksidan, terbukti dapat mencegah disfungsi organ2.
-Penurunan aktifitas OXPHOS (Oxydative Phosphorylation) menyebabkan penuan sel
Akumulasi mutasi mtDNA yg meningkat sejalan dengan bertambahnya umur, juga dapat mempercepat penuaan sel
-Selama penuaan terjadi perpendekan telomer pada berbagai jaringan.Hal ini di modulasi oleh adanya stres oksidatif akibat terakumulasinya radikal bebas di dalam sel.
- Telomer merupakan elemen berulang yg berlokasi pada tiap ujung kromosom dari sel eukariota-Panjang telomer pada manusia
5 – 10 kb
- Fungsi dari telomer adalah untuk menutup ujung kromosom, sehingga mencegah DNA “terbuka”, yang dapat menyebabkan kerusakan DNA, bersatunya kromosom2 dan ketidakstabilan kromosom
- Disebabkan oleh “ end-replication problem” dari DNA polimerase, telomer memendek 50 – 100 pb pada tiap pembelahan sel
-Ketika telomer mencapai panjang yg kritis, menyebabkan fungsi dari telomer tidak optimal. Hal ini menginduksi penuaan sel, yang ditandai oleh adanya gangguan pertumbuhan secara permanent
Tanda-tanda eksternal penuaan:
* Kulit
- kehilangan lapisan lemak dan kelenjar minyak, shg
menjadikan kulit keriput, dan elastisitas menurun.
- sensitif terhadap suhu yang ekstrim
- deposit pigmen melanin
* Rambut
-kehilangan pigmen, dan memutih
* Kuku
- Menebal, karena aliran darah pd jaringan connective tsb berkurang.
