BINDING FORCES BETWEEN MOLECULES
10
In order for molecules to exist in aggregates in gases, liquids, and solids, intermolecular
forces must exist.
An understanding of intermolecular forces is important in the study of pharmaceutical
systems and follows logically from a detailed discussion of intramolecular bonding
energies.
DODY
In order for molecules to exist in aggregates in gases, liquids, and solids, intermolecular forces
must exist.
An understanding of intermolecular forces is important in the study of pharmaceutical systems and follows logically from a detailed discussion of
intramolecular bonding energies.
(1) Repulsive and Attractive Forces
(2) Van der Waals Forces
(3) Ion–Dipole and Ion–Induced
Dipole Forces
(4) Hydrogen Bonds
DODY
Cohesion, or the attraction of like molecules, and adhesion, or the attraction of unlike
molecules, are manifestations of intermolecular forces.
A knowledge of these forces is important for an understanding not only of the properties of
gases, liquids, and solids, but also of interfacial phenomena, flocculation in suspensions, stabilization of emulsions, compaction of
powders in capsules, and the compression of granules to form tablets.
DODY
STATES OF MATTERGases, liquids, and crystalline solids are the three
states of matter. The molecules, atoms, and ions in the solid state are held in close proximity by
intermolecular, interatomic, or ionic forces.
The particles of the solid can oscillate only about fixed positions.
As the temperature of a solid substance is raised, the particles acquire sufficient energy to disrupt the
ordered arrangement of the lattice and pass into the liquid form.
Finally, when sufficient energy is supplied, the molecules pass into the gaseous state.
13DODY
Solids with high vapor pressures, such as iodine and camphor, can pass directly from the solid
to the gaseous state without melting.
This process is known as sublimation, and the reverse process, that is, recondensation to the solid state, may be referred to as deposition.
Certain asymmetric molecules frequently exhibit a fourth phase, more properly termed a mesophase (Greek, mesos, middle), which lies between the liquid and crystalline states. This so-called liquid crystalline state is discussed
later.

Solid dispersions
Solid dispersions: lipophilic drug is incorporated in a hydrophilic matrix
Terjadinya peningkatan laju disolusi
1. Reduksi ukuran partikel 2. Efek solubilisasi dari pembawa 3. Hilangnya sifat agregasi/ agromelasi bhn. aktif hidrofob 4.
<<, laju disolusi >> 2 x x V Log S/ So = ---------------------------- (Kelvin) 2.303 x R x T x r
Kel. kristal mikro (S) meningkat dibanding kel. Kristal makro (So) karena r << Dosis terapi reserpin mikro = 1/3 makro
Sadono
DISPERSI SOLIDA dibedakan menjadi (Chiou & Riegelman, JPS 71; 60 (9) ) :
1. Camp.eutecticum Perband. bhn 1 & 2 tertentu 2. Solid solution 3. Glass solution & glass suspension 4. Endapan amorf dalam pembawa kristalin 5. Pemb. kompleks bhn. aktif & pembawa 6. Kombinasi antar kelima cara
Sadono
TIGA CARA PEMBUATAN SISTIM DISPERSI SOL.
1. Cara/ metoda peleburan 2. idem pelarutan 3. idem peleburan-pelarutan
Sadono
1. Metoda peleburan Melebur campran bhn-bhn pd komposisi eutektikum, kemudian dipadatkan dg. cepat pd penangas es. Gerus dan ayak mesh tertentu Syarat : bhn-bhn dp bercampur sempurna pd. Keadaan melebur dan stabil suhu tinggi Keuntungan : sederhana & ekonomis Kerugian : pengaruh suhu tinggi thd. penguapan, peruraian, perubahan kea- rah polimorf metastabil ke stabil pd. penyimpanan Contoh: Disp. Griseofulvin-pentaeritriol Disp. Spironolakton-PEG 6000
Sadono
2. Metoda pelarutan Melarutkan campuran fisik bhn-bhn pd pelarut organik, diikuti penguapan pelarut pd suhu rendah. Gerus dan ayak mesh tertentu Keuntungan : terhindar peruraian termal Kerugian : pemilihan pelarut yg tidak tok- sik, penghilangan sempurna pelarut, biaya mahal
3. Metoda peleburan pelarutan Melarutkan bhn-bhn obat pd pelarut terpi- lih, diikuti pencampuran pembawa yg telah dilebur Gerus dan ayak mesh tertentu Kerugian : terbatas obat terapi rendah
Sadono
Persyaratan pembawa : 1. mudah larut air & laju dis. tinggi 2. tidak toksik 3. inert secara farmakologik 4. dp bercampur secara kimia dl keadaan pa- dat dan tidak membentuk ikatan kom- pleks yg kuat dengan bhn aktif 5. stabil pd suhu tinggi (met. peleburan) 6. larut pd pelarut organik (met. pelarutan) Contoh pembawa : PEG 4000/ 6000 s/d 20000, PVP K-25 K-30, K-90, Laktosa, sugars, as. Sitrat, as. suksinat, urea.
Apa perbedaan camp.fisik dan disp.solida ? Pd campuran fisik internal structure crystal masing-maing bahan tetap

Campuran fisik dan dispersi solida hanya terbedakan pd keadaan solida/ bukan li-
kuida atau lainnya. Why ?
Sadono
A suitable powder dispersion as the basis of preparing a wide range of very import- ant pharmceutical dosage forms such as capsules, granulations, compressed ta- blets and throat lozenges.
Tablets and capsules can then be treated with special coatings, often a liquid poly- meric dispersion, to improve the stability or release of the drug, or for identification purposes.

Microemulsions (1)
• Literature: confusing/contradictive
• Consist of: water / oil / surfactant / co-surfactant
• Something between a micelle and an emulsion
• Swollen or large micelle
• Size: - micelle: 5 - 10 nm
- microemulsion: 10-200 nm
- emulsion: > 1000 nm
• Optically isotropic because particle size is smaller
than wavelength visible light (emulsions are turbid)
• Thermodynamically stable (emulsions are not)
• Lipophilic drugs can be dispersed molecularly
Microemulsions (2)
Co-surfactant (usually an alcohol, e.g. pentanol)
Interfacial tension is close to zero
Microemulsions are formed spontaneously
Microemulsions are thermodynamically stable
SMEDDS: SELF EMULSIFYING DRUG DELIVERY SYSTEMS
Nanosuspensions
Saturation concentration around small particles is higher than around large particles
solubility especially strongly increases when particle size < 1 m
Top down methods:
- Dry milling: particles smaller than 1 m not possible
Wet milling e.g. high pressure homogenization
Bottom up methods:
- Controlled crystallization/precipitation
Disadvantage:
- Large amounts of surfactants are required to
prevent aggregation and Ostwald ripening
Complexing agents (cyclodextrins)
+
CD + lipophilic drug complex
lipophilic cavity hydrophilic outside
-CD -CD -CD
Molecular weight 972 1135 1297
Glucose monomers 6 7 8
Internal cavity diameter (nm) 0.5 0.6 0.8
Three types of cyclodextrins (+ derivatives)
Fusion method: - heat until carrier and drug mix (e.g. extruder)
- rapid cooling
Disadvantages:
- limited miscibility during heating
- thermal degradation
- phase separation during cooling can easily occur
Current methods to preparesolid dispersions (1)
Solvent method:
- dissolve carrier and drug in one solvent (mixture)
- evaporation of solvent
(e.g. vacuum drying, spray drying)Disadvantages:
- phase separation can easily occur
- limited choice of solvent (mixtures)
(usually class II solvents e.g. chloroform)
- limited choice of matrix (e.g. PVP and PEG)
Current methods to preparesolid dispersions (2)
Current methods to prepare solid dispersions (3)
Alternative solvent method: - dissolve carrier and drug in one solvent (mixture) - solvent is removed by freeze drying
Freeze drying: - solution is frozen - pressure is reduced - solvent is removed by sublimation
Sublimation: - evaporation of a substance from the solid state without passing the liquid state
